Different from international“stool culture medium”, Chinese“stool culture medium” just refers to the culture of Salmonella and Shigella.
Although Salmonella and Shigella may be isolated from stool samples directly inoculated on the plate, Salmonella and Shigella can be isolated to the maximum extent by inoculating the stool samples in zengmei broth. However, Salmonella enrichment broth is often highly selective, which can inhibit some serotypes of Salmonella, especially Salmonella typhoid serotype; there is no reliable enrichment medium for Shigella. Gram negative broth and selenite sel broth are commonly used to isolate Salmonella and Shigella from stool.
In order to obtain the best separation effect of Salmonella and Shigella, the selective plate should be applied. The low selective medium includes MAC agar and eosin methylene blue agar. Medium selective medium includes XLD, sodium deoxycholate citrate agar, SS agar, he agar, intestinal medium of National Institute of serum and Salmonella comaga agar. The highly selective medium includes bismuth Sulfite Agar (suitable for isolating Salmonella typhoid serotype) and bright green agar. Salmonella agar should be used with caution as it inhibits the growth of some type-1 strains of Shigella dysenteriae.
